• Santhanam was still a TV comedian when Jyothirmayi, already a known face, agreed to star opposite him in Idhaya Thirudan (2006). She later said she took the gamble because his comic timing in the audition made her laugh so hard she couldn't say no.
• In Arai En 305-il Kadavul (2008), Jyothirmayi played the straight woman to Santhanam's manic energy. She deliberately underplayed every reaction, letting his rapid-fire dialogue land harder. He later admitted her stillness forced him to sharpen his delivery.
• The Telugu remake Gadi No.305 lo Devudu (2008) was shot back-to-back with the Tamil original. Santhanam and Jyothirmayi re-shot every scene in Telugu themselves, making them one of the rare Tamil-Telugu bilingual comedy pairs of that era.
• During the shoot of Idhaya Thirudan, Santhanam would run his lines past Jyothirmayi before every take. She became his unofficial script doctor, cutting jokes she thought were too crass. He trusted her judgment so much that he never argued once.
• Jyothirmayi told a Chennai press meet in 2008: 'Santhanam made me laugh so much on set that the director had to cut the shot. He doesn't just act funny — he lives funny.'
