• Their first film together, Super Police (1994), almost didn't happen. Jayasudha was hesitant to play a mother to a grown-up hero so early in her career. Kota Srinivasa Rao personally convinced her, saying the role had more depth than a typical 'amma' part.
• In Seethamma Vakitlo Sirimalle Chettu (2013), Kota Srinivasa Rao played the father and Jayasudha played the mother. The director said Kota would deliberately flub his lines during rehearsals just to make Jayasudha laugh, and that natural, unscripted laughter made it into the final cut.
• Kota Srinivasa Rao and Jayasudha had a running joke on every set: whoever finished their shot first had to bring the other a cup of filter coffee. This started during Yuvakudu (2000) and continued through Maharshi (2019).
• Their pairing as the elderly parents in Seethamma Vakitlo Sirimalle Chettu (2013) directly inspired the casting of veteran actors as the romantic leads' parents in several subsequent family dramas, including the 2016 hit 'Srirastu Subhamastu'.
• Jayasudha once said in an interview: 'Kota garu is the only co-star who can make me forget my lines because I'm laughing so hard. He's a menace on set, but a sweet menace.'
• In Ooranthaa Anukuntunnaaru (2019), their only hit together, Kota Srinivasa Rao improvised a scene where he scolds Jayasudha's character for being too soft on their son. She responded by silently tearing up — a reaction he didn't expect. The director kept the camera rolling and used that single take.
