• Brahmanandam personally recommended L.B. Sriram for the role of 'Bobbili Pasupathi' in Bobby (2002). Sriram was a lesser-known comedian at the time, and that role became his breakout comic character in Telugu cinema.
• In Ninne Premista (2000), Brahmanandam played the straight man to L.B. Sriram's over-the-top buffoonery. Sriram would improvise gibberish dialogues on set, and Brahmanandam would react with deadpan silence — a rhythm they reused in five of their eight films together.
• L.B. Sriram once said in an interview that Brahmanandam would call him before every shoot to ask, 'Emanna kotha comedy undha?' (Any new comedy ideas?). They would then rehearse their scenes in Brahmanandam's car for 20 minutes before entering the set.

• In Daruvu (2012), their only film together after a 9-year gap, Brahmanandam deliberately slowed down his delivery to match Sriram's stammering comic style. The director later said that Brahmanandam insisted on reshooting a scene because he felt Sriram's timing was off — and the final take became the film's most-quoted comedy bit.
