• Their first film together, Vamsi (2000), was a flop. But director K. Raghavendra Rao saw their timing in the outtakes and insisted they be paired again in Jayam Manade Raa the same year.
• In Manmadhudu (2002), Brahmanandam played a stammering sidekick and Jaya Prakash Reddy played the villain. Brahmanandam would deliberately flub his lines to make Jaya Prakash Reddy break character — and the director kept those takes because they were funnier.
• Their comedy track in Golmaal (2003) — where Brahmanandam's character keeps mispronouncing Jaya Prakash Reddy's name — became a meme template in Telugu college hostels for years. It directly inspired a similar gag in the 2010 film Maryada Ramanna.
• On the sets of Apparao Driving School (2004), Jaya Prakash Reddy would bring homemade pickles from his village for Brahmanandam. They shared lunch together every single day of that shoot — a ritual they kept up for every film after.
• Brahmanandam once said in a 2016 interview: 'Jaya Prakash Reddy garu is the only actor who can make me laugh while I'm acting. I'd be in the middle of a scene and he'd do something with his eyes — I'd lose it.'
